Single-cell analyses can reveal new levels of complexity of biological systems. The ability to measure the heterogeneity within a system at a single-cell resolution has led to the discovery of new cell types and expanded our understanding of the diverse regulatory pathways and states of individual cells. However, when starting from solid tissues, manual methods to dissociate them reproducibly into high-quality single-cell or nuclei suspensions may be challenging, as tissue-specific protocols may need to be established, be laborious, and with variable yields and viability. Here, we demonstrate the benefits of employing automated solid tissue dissociation into single-cell and -nuclei suspensions from a variety of tissue types. They include enhances in efficiency, reproducibility, and yield, while minimizing manual processing time and potential biases. Through case studies, we discuss the benefits of cell or nuclei isolation depending on application needs, as well as how automated isolation methods may improve sequencing results over manual methods.
